摘要:

目的 探討小鼠骨折模型的建立難點(diǎn)、術(shù)式特點(diǎn),分析模型建立后不同時(shí)相的骨折愈合特點(diǎn),確定更具優(yōu)勢(shì)的小鼠骨折模型。方法 以C57BL/6野生型小鼠為平臺(tái)建立脛骨開放橫形骨折髓內(nèi)固定模型及股骨閉合橫形骨折髓內(nèi)固定模型。術(shù)后連續(xù)觀察并在術(shù)后第4、7、10、14、21、28、35天收取骨折部位標(biāo)本,采用X線、micro-CT、組織形態(tài)計(jì)量學(xué)等方法對(duì)上述骨折模型進(jìn)行比較分析,并連續(xù)觀察小鼠活動(dòng)能力。結(jié)果 開放性脛骨骨干橫形骨折模型單批建立成功率為94.2%,無新發(fā)現(xiàn)骨折,無固定針移位現(xiàn)象。閉合性股骨骨干橫形骨折模型單批建立成功率為80.0%,17.9%的小鼠發(fā)現(xiàn)粉碎性骨折或手術(shù)區(qū)域外骨折,14.3%出現(xiàn)固定針向近端移位現(xiàn)象。脛骨骨折組小鼠骨性骨痂體積百分比及骨連接密度在術(shù)后第10、14、21及28天均顯著高于股骨骨折組小鼠(P<0.05)。術(shù)后第7天,脛骨骨折組小鼠軟骨骨痂面積百分比顯著高于股骨骨折組小鼠;術(shù)后第10、14、21、28天,脛骨骨折組小鼠骨性骨痂面積百分比顯著高于股骨骨折組小鼠(P<0.05)。脛骨骨折組小鼠和股骨骨折組小鼠的軟骨骨痂面積百分比分別在術(shù)后第7天和第10天到達(dá)峰值。術(shù)后脛骨骨折組小鼠活動(dòng)能力的恢復(fù)快于股骨骨折組小鼠。結(jié)論 小鼠開放性脛骨骨干橫形骨折髓內(nèi)針固定模型是較為理想的小鼠骨折模型。

Objective To study the key points and operative characteristics of establishment of the bone fracture models in mice and to analyze and compare the fracture healing characteristics of two chosen fracture models in mice in different time points post-operatively,in order to definite the advantageous fracture model in mice.Methods An open transverse tibial fracture with intramedullary needle fixation model and a closed transverse femoral fracture with intramedullary needle fixation model were selected,which were both established in C57BL/6 wild-type mice.The characteristics of bone fracture healing of these mice were serially analyzed and compared by X-ray,micro-CT and histomorphometry.The observations of mobility were also included.Time points were set by day 4,7,10,14,21,28 and 35 post-operatively.Results The one batch successful rate of model establishment was 94.2% on tibiae;no other fracture and displacement of fixation needle were observed.The one batch successful rate of model establishment was 80.0% on femurs;comminuted fractures or fractures outside the operation region were observed in 17.9% of mice and 14.3% of the fixation needles were found displaced proximally.Callus mineralized volume fraction and connectivity density of calluses on tibiae were significantly higher in comparison to calluses on femurs by day 10,14,21 and 28 postoperatively (P<0.05).A larger cartilage area was observed by day 7 and a larger bone area was observed by 10,14,21 and 28 postoperatively in calluses on tibiae when compared to calluses on femurs (P<0.05).The cartilaginous callus fraction reached peak level in tibial tissues by day 7 and in femoral tissues by day 10 postoperatively.The recover period from fracture in terms of mobility was shorter in mice operated by tibial fracture.Conclusions Comparing the two animal fracture models,the open transverse tibial fracture with intramedullary needle fixation model is better for the studies of bone fracture healing.
