摘要:

目的 研究機(jī)械牽張作用對(duì)大鼠小梁網(wǎng)細(xì)胞蛋白組學(xué)的表達(dá)影響。方法 實(shí)驗(yàn)對(duì)象選用對(duì)數(shù)生長(zhǎng)期的大鼠小梁網(wǎng)細(xì)胞。采用FX-5000牽張應(yīng)力加載系統(tǒng)拉伸細(xì)胞, 提取細(xì)胞全蛋白后進(jìn)行基于鳥(niǎo)槍法的質(zhì)譜測(cè)試和蛋白質(zhì)的鑒定。運(yùn)用微陣列顯著性分析 (significance analysis of microarray, SAM) 方法分析實(shí)驗(yàn)組和對(duì)照組的蛋白組學(xué)差異;對(duì)差異蛋白進(jìn)行基于Gene Ontology (GO) 的功能注釋、富集和Kyoto Encyclopedia of Genes and Genomes (KEGG) 通路富集并篩選出顯著富集的蛋白。結(jié)果 SAM法篩選出差異蛋白共計(jì)57, 均表達(dá)上調(diào), 分布在細(xì)胞各個(gè)區(qū)域, 參與細(xì)胞發(fā)育、免疫調(diào)節(jié)、刺激反應(yīng)、代謝、細(xì)胞黏附等過(guò)程。其中24種顯著富集在9個(gè)隸屬生物學(xué)過(guò)程的GO term, 進(jìn)一步KEGG通路富集分析顯示熱休克蛋白和核糖體蛋白顯著富集在內(nèi)質(zhì)網(wǎng)蛋白加工通路和核糖體通路中。結(jié)論 機(jī)械牽張對(duì)大鼠小梁網(wǎng)細(xì)胞的蛋白質(zhì)表達(dá)影響區(qū)域廣, 過(guò)程復(fù)雜。富集分析說(shuō)明熱休克蛋白和核糖體蛋白在牽張刺激下的應(yīng)答保護(hù)和眼壓調(diào)控中起到了重要作用。

Objective To study differential proteomics analysis of rat trabecular meshwork cells under mechanical stimulation. Methods The rat trabecular meshwork cells in logarithmic growth period were selected.Mechanical stretching was loaded by FX-5000 tension system. The total proteins were extracted and identified by shotgun-based mass spectrometry test and analysis. Differential expressed proteins between sample group and control group were analyzed by significance analysis of microarray (SAM) method. Gene Ontology (GO) annotation, GO enrichment and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment were used to gain significantly enriched differential proteins.Results A total of 57 differential proteins were screened by SAM method, all of which were up-regulated and distributed in various regions of the cell, and were involved in the processes of cell development, immunomodulation, stimulus response, metabolism and cell adhesion. Twenty-four of them were significantlyenriched in 9 biological process GO terms and 2 KEGG pathways, mainly assigned to heat shock proteins and ribosome proteins. Conclusions The effect of mechanical strain on protein expression of rat trabecular meshwork cells was wide and complex. The enrichment analysis showed that heat shock proteins and ribosome proteins played an important role in the response protection and intraocular pressure regulation under mechanical strain.
