水性組織透明化技術的研究進展與應用

摘要:

由于生物樣本的不透明性,以及組織深入成像的清晰度和深度不佳,因此大型、厚組織3D成像技術的發(fā)展與創(chuàng)新成為近年來的研究熱點。組織透明化機技術是通過各種物理或化學的方式將組織變透明,并結合熒光顯微鏡來實現組織或器官三維成像的新技術,因其良好的熒光保存及透明效果,在軟組織領域,尤其是神經領域得到了越來越廣泛的使用。近年來,各種新型的組織透明化技術不斷被報道出來,本文就被動浸潤法水性透明化技術、水化法水性透明化技術、水凝膠嵌入法水性透明化技術的研究進展及應用做一綜述。

The development and innovation of 3D imaging technology for large and thick tissues have become a research hotspot in recent years due to the opacity of biological specimens and the lack of clarity and depth of tissue sample imaging. The hydrophilic reagent-based tissue clearing technology, through a variety of physical or chemical methods to clear tissues, enables the three-dimensional imaging of tissues or organs with fluorescence microscopes, which has been widely adopted to clear soft tissues, especially nerves because of its satisfactory fluorescence preservation and clearing effects. An increasing number of new tissue clearing techniques have constantly been reported in the last few years. In this paper, the research progress and application of passive infiltration water - based transparency technology, hydration water transparency technology and hydrogel embedding technique for waterborne transparency will be reviewed.
