摘要:

目的 建立一種血管內(nèi)皮生長(zhǎng)因子(vascular endothelial growth factor,VEGF)在視網(wǎng)膜內(nèi)異常表達(dá)情況的可視化和定量評(píng)估方法,為糖尿病視網(wǎng)膜病變(diabetic retinopathy, DR)的早期檢測(cè)和治療提供幫助。方法 對(duì)大鼠視網(wǎng)膜鋪片進(jìn)行免疫熒光染色和成像。對(duì)VEGF和微血管病變區(qū)域進(jìn)行分割、提取與三維重建,采用VEGF陽(yáng)性單位、微血管密度、分形維數(shù)等定量參數(shù)對(duì)結(jié)果進(jìn)行評(píng)估,比較糖尿病視網(wǎng)膜病變8周模型組(n=10)與正常對(duì)照組(n=10)的差異。結(jié)果 在DR病變模型中,VEGF探針高表達(dá)區(qū)域與血管異常區(qū)域存在共定位,VEGF聚集于視網(wǎng)膜微血管分叉或膨大的血管壁表面。DR組的微血管分形維數(shù)、密度和三維血管彎曲度明顯高于正常對(duì)照組(P<0.05);VEGF陽(yáng)性單位(positive unit, PU)值在DR組中明顯高于對(duì)比組(P<0.05);微血管空隙度差異無(wú)統(tǒng)計(jì)學(xué)意義(P>0.05)。結(jié)論 本研究提出一種VEGF定量評(píng)估方法,能夠定性和定量地反映VEGF在DR模型視網(wǎng)膜中的異常分布,為抗VEGF藥物研究提供定量及可視化手段。

Object To establish a method for visualizing and quantifying the abnormal expression of vascular endothelial growth factor (VEGF) in the retina ,and to help the early detection and treatment of diabetic retinopathy (DR). Methods Immunofluorescence staining and imaging were performed on the rat retinal flatmounts. VEGF and microvessels were segmented, extracted and reconstructed. Quantitative parameters such as positive VEGF units, microvascular density and fractal dimension were used and the differences between the 8-week diabetic retinopathy model group (n=10) and the normal control group (n=10) were compared. Results In the DR model, there was colocalization between the high expression region of VEGF probe and the abnormal region of vascular, and VEGF accumulates on the surface of retinal microvascular bifurcation or swollen vessel wall. The fractal dimension, density and curvature of microvessels in DR group were significantly higher than those in normal control group (P<0.05). The PU value of VEGF in DR group was significantly higher than that in comparison group (P<0.05). There was no significant difference in microvascular void (P>0.05). Conclusions In this study, a quantitative evaluation method of VEGF is proposed, which can reflect the abnormal distribution of VEGF in the retina of DR model qualitatively and quantitatively, and provide a quantitative and visual way for the research of anti-VEGF drugs.
