摘要:

目的 研究不同濃度的銀納米顆粒對神經(jīng)元毒性的劑量-效應(yīng)關(guān)系,探索銀納米顆粒對神經(jīng)元的毒性機理。方法 首先培養(yǎng)一種活性好、生長狀態(tài)優(yōu)良的原代神經(jīng)元,將不同濃度(2.5~500μg/mL)的銀納米顆粒加入神經(jīng)元中作用24h后,通過MTT法計算細胞增殖率,并分析不同濃度的銀納米顆粒對神經(jīng)元毒性的劑量-效應(yīng)關(guān)系。結(jié)果 在25~250μg/mL范圍內(nèi),細胞數(shù)量與形態(tài)呈現(xiàn)不同的變化,銀納米顆粒濃度與細胞增殖率呈負相關(guān)。結(jié)論 在25~250μg/mL的濃度范圍內(nèi),銀納米顆粒對神經(jīng)元細胞的毒性呈劑量-效應(yīng)關(guān)系。

Objective To study the dose-effect relationship between silver nanoparticles with different concentrations and the neurotoxicity, and also explore the mechanism of silver nanoparticles to neurotoxicity.  Method A group of primary neurons in excellent growth status was cultured, and then contacted with different concentrations (2.5-500μg/ml )of silver nanoparticles for 24h. After that we calculated the cell proliferation rate by MTT test and analyzed the dose-effect relationship between the concentrations of silver nanoparticle with neurotoxicity. Result Within the silver nanoparticle concenrtation of 25-250μg/ml, both cellular mount and morphology changed, and it was showed anticorrelation between silver nanoparticle concentration and cell proliferation rate. Conclusion Within the sliver nanoparticles concenrtation of 25-250μg/ml, silver nanoparticles shows a dose-effect relationship on neurotoxicity.
