摘要:

目的 探討ssDNA單層膜的制備的最佳條件。方法 利用自組裝單分子膜技術(shù),將人工合成并修飾的含22個堿基的特異性單鏈DNA序列固定到金電極上制備成DNA修飾的金電極,通過控制DNA溶液的濃度、作用時間,以及改變修飾基團(tuán)探討最佳的修飾條件,并利用循環(huán)伏安法初步研究了該修飾電極的電化學(xué)行為。結(jié)果 通過對不同標(biāo)記探針、不同探針濃度及不同標(biāo)記時間的表征,結(jié)果顯示巰基化ssDNA和二硫鍵修飾的ssDNA探針濃度為80μmol /L,自組裝固定時間為12 h時對DNA探針的固定效率最佳。結(jié)論 巰基化ssDNA和二硫鍵修飾的ssDNA通過自組裝方法可在金電極表面形成穩(wěn)定、有序的自組裝單分子膜(self-assembled monolayer,SAM),可應(yīng)用于SAM為基底的DNA傳感器的設(shè)計與應(yīng)用。

Objective To investigate the best preparation condition of ssDNA single-layer film. Methods A DNA modified gold electrode was obtained with the technology of self-assembled monolayer(SAM). The concentration of DNA and the time of modification were studied by cyclic voltammetric technique. Results The results showed that a SAM was formed on the surface of gold electrode. The best SAM could be obtained in 80μmol/L capture fixed for 12 h. Conclusions The generation of sulfur gold key could form a stabilized SAM in gold electrodes.
