摘要:

目的 探討海藻酸鈉支架復合組織工程化軟骨細胞對兔佐劑性關節(jié)炎膝關節(jié)軟骨缺損修復的影響。方法 將新西蘭白兔分為正常對照組、佐劑性模型組和海藻酸鹽支架治療組。模型組和治療組兔膝關節(jié)內注入弗氏佐劑建立佐劑性關節(jié)炎模型。抽取治療組兔左后健側肢體的骨髓,用Percoll法分離出自體骨髓間充質干細胞(ABMSC),體外培養(yǎng)傳代至P3代并誘導分化為軟骨細胞。將軟骨細胞與海藻酸鈉支架混合注入治療組的病變膝關節(jié)。對照組和模型組關節(jié)注射等量的生理鹽水。海藻酸鈉支架復合組織工程化軟骨細胞治療1個月后處死兔,應用Ⅱ型膠原免疫組織化學法染色和HE染色,觀察各組兔的膝關節(jié)關節(jié)軟骨缺損的修復情況。結果 誘導成功軟骨細胞且生長良好,呈現明顯的紡錘體形,細胞成活率達98.5%。軟骨細胞與海藻酸鈉復合后體外培養(yǎng)顯示細胞生長旺盛,增殖成株狀或島狀,株狀細胞團周圍有類似的軟骨陷窩形成,細胞排列密集,核圓形。組織形態(tài)學結果顯示,治療組與模型組比較,治療組關節(jié)炎癥反應明顯減輕,關節(jié)Ⅱ型膠原陽性軟骨細胞顯著增高,軟骨缺損得到明顯修復。結論 海藻酸鈉支架復合組織工程化軟骨細胞對兔佐劑性關節(jié)炎膝關節(jié)軟骨缺損有一定的修復作用。
 

Objective To explore the effect of cartilage defect repair by using engineered chondrocyte composite with bone marrow stem cells (BMSCs) cultured on the sodium alginate scaffold in adjuvant-induced arthritis (AA) rabbits. Methods Twenty-four New Zealand white rabbits were divided into control group,model group and alginate treatment group at random. Complete Freund’s adjuvant was used to induce adjuvant-induced arthritis in model group and alginate treatment group. Bone marrow of the rabbits’ healthy left hindlimbs was collected from the alginate treatment group,autologous bone marrow mesenchymal stem cells(ABMSCs) were isolated by the method of Percoll. The third generation (P3) ABMSCs were cultured and differentiated into chondrocytes. Engineered chondrocytes composite with sodium alginate scaffold were injected in the alginate treatment group,and the control group and model group were injected with saline in rabbits’ articulus. Rabbits were sacrificed after one month of the treatment. Repair of articular cartilage defects were observed by HE and type Ⅱ collagen immunohistochemical staining in rabbits’ articular tissue. Results Engineered chondrocytes grew well and showing spindle-shaped,cell survival rate was 98.5%. Chondrocytes with sodium alginate composite also grew well in vitro,appearing plant-like or island,similar cartilage lacuna formed around the cells. The cells arranged densely,nucleus showing round. Compared with model group,histomorphometric results showed that inflammation was significantly reduced and type Ⅱ collagen positive cells remarkably increased in alginate treated rabbits. The repair of cartilage defect was significantly improved in alginate treatment group. Conclusions Engineered chondrocytes composite with alginate scaffold can repair cartilage defect in adjuvant-induced arthritis rabbits.
