摘要:

目的 分析晚期糖基化終產(chǎn)物受體(receptor for advanced glycation endproducts,RAGE)的遺傳變異

與中國漢族人群高血糖的關(guān)系。方法 調(diào)查對(duì)象來自江蘇省農(nóng)村地區(qū)14 469人群中,選取18~62歲成年對(duì)象

共2 731人進(jìn)行空腹血糖水平流行病學(xué)調(diào)查;收集流行病學(xué)資料和體格檢查資料并檢測血糖等臨床生化指標(biāo)

。位點(diǎn)選擇采用連鎖不平衡分析的方法,選取RAGE基因及其上游的3個(gè)tagSNP rs1800624、rs204993、

rs184003進(jìn)行基因分型和關(guān)聯(lián)分析。結(jié)果 校正年齡、性別、體力活動(dòng)和BMI混雜因素前后,rs1800624、

rs204993和rs184003位點(diǎn)與血糖水平的關(guān)聯(lián)均無統(tǒng)計(jì)學(xué)意義。不同基因型之間血糖水平比較也未發(fā)現(xiàn)上述3

個(gè)SNP變異與血糖數(shù)量性狀存在關(guān)聯(lián),進(jìn)一步按性別因素進(jìn)行分層分析,結(jié)果顯示,在男性人群中,病例組

rs184003位點(diǎn)的不同基因型與血糖水平關(guān)聯(lián)有統(tǒng)計(jì)學(xué)意義,OR(95%CI)為0.212~2.261,P值為0.01;校正年

齡、體力活動(dòng)指數(shù)和BMI混雜因素后,結(jié)果仍有統(tǒng)計(jì)學(xué)意義,OR(95%CI)為0.064~2.305,P值為0.002。結(jié)論

未發(fā)現(xiàn)RAGE基因rs1800624、rs204993和rs184003位點(diǎn)的變異與血糖水平存在顯著關(guān)聯(lián)。
 

To evaluate the association between three single nucleotide polymorphisms of

receptor for advanced glycation endproducts gene and hyperglycemia in Han Chinese

population.Methods Totally 2 731 adult subjects aged 18-62 were recruited from a rural

population of 14 469 subjects in Jiangsu Province by an epidemiological stratification

sampling approach and epidemiological data and serum glucose were collected and

detected.Linkage disequilibrium (LD) analysis were applied and three tagging single nucleotide

polymorphisms (tagSNPs) of RAGE gene (rs1800624,rs204993,rs184003) were selected for

genotyping and analyzing for the association with fasting blood glucose level.Results Further

adjusted for confounding factors including age,sex,physical activity index and body mass

index,no statistical significant difference was detected in the distribution of genotypes for

SNP rs1800624,rs204993 and rs184003 between cases and controls.And there was no significant

glucose difference among genotype variations of rs1800624,rs204993 and rs184003.Further

stratification analysis by sex showed that there was statistical significant difference of

glucose level among genotype variations of SNP rs184003 in male.Conclusions Our result

suggests that SNP rs1800624,rs204993 and rs184003 of RAGE gene are not associated with

increased risk of hyperglycemia in Han Chinese men and further replication study would be

warranted.
